DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Glyceryl trinitrate [55-63-0] and pentaerythritol tetranitrate [78-11-5] administered orally to humans were rapidly metabolized, and the peak blood concn. of glyceryl trinitrate metabolites, 1,2-glyceryl dinitrate [621-65-8] and 1,3-glyceryl dinitrate [623-87-0], appeared at 15 and 180 min. postadministration. The max. 78-11-5 and 621-65-8 are cas registry numbers of chemicals which are used as reagents here. level of 1-glyceryl mononitrate [624-43-1] and 2-glyceryl mononitrate [620-12-2] appeared at 120 min. Similar kinetics were obsd. with pentaerythritol dinitrate [1607-01-8] and mononitrate [1607-00-7]. A large part of the nitrate metabolites was excreted in urine as conjugates. The total excretion during the 1st 24 h accounted for 22% of the administered dose in the case of glyceryl trinitrate. In total, 19% of the dose of pentaerythritol trinitrate was found as nitrate metabolite in urine within 24 h. A method using gas-chromatog. is described for the detn. of glyceryl trinitrate and pentaerythritol tetranitrate in blood and urine. .
