DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) The effects of strychnine (I) [57-24-9] bicuculline [485-49-4], picrotoxin [124-87-8], and L-allylglycine [16338-48-0] on the transsynaptic destruction of medullary dorsal horn neurons were examd. following transection of the inferior alveolar nerve in adult rats. Strychnine and L-allylglycine enhanced the transsynaptic effect of nerve transection and caused degeneration of many dorsal horn neurons, whereas bicuculline and picrotoxin did not. The removal of glycinergic and GABAerigic postsynaptic inhibition appears to enhance the transsynaptic destructive activity which follows the peripheral nerve transection.

Both isomers inhibited brain glutamate decarboxylase (EC 4.1.1.15) (I) [9024-58-2] to a similar extent in vitro, but the L-isomer was the stronger inhibitor in vivo. Allylglycine concns. that inhibited I in vivo were ineffective in vitro. Allylglycine oxidn. products of L- or D-amino acid oxidase action inhibited I in vitro. A common intermediate (possibly 2-keto-4-pentenoic acid) may be responsible for the in vivo I inhibition and the induction of convulsions.
