DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) l-Stepholidine (I) [16562-13-3] caused contralateral circling in rats with unilateral nigral lesions induced by 6-hydroxydopamine (6-OHDA) or electrocoagulation. In 6-OHDA-lesioned rats I potentiated apomorphine-induced contralateral circling, but reversed amphetamine-induced ipsilateral rotation. In electrolytically lesioned rats, both apomorphine- and amphetamine-induced ipsilateral rotations were reversed by I. Apparently, I is a dopamine antagonist with some agonistic activity.

They were characterized by m.p. detns., TLC, UV and IR spectrometry, NMR, and mass spectrometry. Nine of them are known alkaloids, i.e., l-tetrahydropalmatine (I) [483-14-7], l-corydalmine [30413-84-4], l-stepholidine [16562-13-3], l-discretamine [1356-73-6], d-stepharine [2810-21-1], stepharanine [17369-30-1], jatrorrhizine [3621-38-3], palmatine [3486-67-7], and dehydrocorydalmine [6877-27-6]. The other one is a new alkaloid and is named dehydrodiscretamine (II) [89560-99-6]. Among them, l-stepholidine can suppress pain and retard brain dopaminergic receptor function.
