DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Evaluation of bis[1,2-bis(diphenylphosphino)ethane]gold(I) chloride (I) [19624-67-0] in mice bearing i.p. P388 leukemia demonstrated that the compd. produced an av. of 87% increase in life span at its maximally tolerated dose (2-3 mmol/kg/day for 5 days). Activity was also seen in i. 19624-67-0 and 15663-27-1 which are cas registry numbers are also used here.p. M5076 reticulum cell sarcoma (60% increase in life span) and s.c. mammary adenocarcinoma 16/c. Modest activity was evident in i.p. B16 melanoma and L1210 leukemia. A subline of P388 leukemia resistant to cisplatin [15663-27-1] was not cross-resistant to I. In addn., combination therapy of I and cisplatin against i.p. P388 demonstrated an advantage over single-agent therapy. In vitro studies of I showed that the compd.: (a) is cytotoxic to tumor cell lines; (b) is only minimally inhibited in its cytotoxic activity by the presence of serum; (c) produces DNA protein cross-links and DNA strand breaks in cells; and (d) inhibits macromol. synthesis with a preferential inhibitory effect on protein synthesis relative to DNA and RNA synthesis. 31P NMR spectroscopy indicated that the compd. is stable in the presence of serum proteins, thiols, or disulfides and that it reacts with Cu(II) resulting in the formation of a Cu(I)-bis(diphenylphosphino)ethane complex [1663-45-2]. The results of these in vivo and in vitro expts. suggest that the contrasting pharmacol. profile of I with respect to other gold(I) phosphine complexes may be related to both the kinetic stability of the complex and its stability in the presence of thiols. .

Unstable complex 2d (P-P = dmpe) was generated in situ. X-ray structures are reported for 1e and 2e-g. Treatment of 2a-g with CO in CH2Cl2 at -90° yields the (P-P)Pd(CH3)(CO)+ complexes 3a-g. Barriers to migratory insertion in 3a-g were detd. with the ordering to be: 3f (dippp) u 3g (dppb) < 3e (dppp) ? 3a (dppee) u 3b (dpbz) u 3c (dppe) < 3d (dmpe). Exposure of 2a-g to ethylene at -80° yields the ethylene complexes (P-P)Pd(CH3)(C2H4)+ (5a-g). Barriers to migratory insertion in these complexes were detd. by NMR spectroscopy to be: 5b (dpbz) u 5e (dppp) u 5f (dippp) u 5g (dppb) < 5a (dppee) u 5c (dppe) < 5d (dmpe). Complexes (P-P)Pd(CH2CH3)(C2H4)+ (6a-e,g) produced from 5a-e,g under C2H4 are catalyst resting states for the dimerization of C2H4 to butenes. In the case of 5f (P-P = dippp), the catalyst resting state produced is the b-agostic Et complex (dippp)Pd(CH2CH2-m-H)+ (8f), which has been isolated. This complex exhibits two dynamic processes studied by VT-NMR:interchange of Ca and Cb (DGG = 10.3(2) kcal/mol) and rotation of the agostic Me group (DGG ca. 6.4 kcal/mol). The b-agostic Pr complex 7f has been generated and identified as the Pr isomer (dippp)Pd(CH2CH2-m-HCH3)+.
