DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) Human kidney cortical and medullary cells were incubated in primary culture as an exptl. model in order to compare the effect of prostaglandin E2 [363-24-6] on the adenylate cyclase [9012-42-4]-cyclic AMP [60-92-4] system at a renal level with the effects of parathormone [9002-64-6] and desamino1-D-arginine8-vasopressin [16679-58-6]. Prostaglandin E2 is a more potent stimulator of cyclic AMP accumulation in cortical cells than in medullary ones, the minimal ED being 3 nM in the cortex and 30 mM in the medulla. The maximal response was obtained with 30 mM in cortical cells and with 0.3 mM in medullary ones. The effects of combining submaximal and maximal active doses of the 3 hormones were always additive when tested on cortical cells. However, prostaglandin E2, at doses effective on the medullary cell adenylate cyclase system, produced an increase of cyclic AMP accumulation significantly lower than expected when incubated with submaximal and maximal amts. of desamino1-D-arginine8-vasopressin. Thus, distinct receptors for parathormone, arginine-vasopressin, and prostaglandin E2 exist at the cortical level; the possible role of prostaglandin E2 in the counter-regulation of arginine-vasopressin action on medullary cells is unlikely, in view of the high prostaglandin E2 concn. which is necessary to exert this effect.

DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) Prostaglandin (PG) prodn. by the kidney is known to be reduced both in vivo and in vitro in rats with hereditary diabetes insipidus (DI), totally lacking ADH [11000-17-2]. Exogenous ADH (using dDAVP [16679-58-6]) restores normal PG excretion in these rats. On the other hand, osmolality in vitro, and urine flow rate in vivo have been shown to influence PG synthesis rate. In order to det. whether the decreased PG synthesis of DI rats is due to the lack of ADH itself or to low tissue osmolality, in vivo and in vitro PG prodn. was studied in DI rats in which urine osmolality had been raised either with ADH or without ADH (by dehydratation) and in control DI rats. ADH administration and dehydration led to similar urine osmolalities (900-1,000 milliosmoles/kg H2O vs. 150 in controls). Only ADH administration (but not dehydration) increased PG urinary excretion and subsequent in vitro papillary synthesis. Thus, ADH increases PG synthesis by the renal papilla directly and not through its effects on papillary osmolality.
