DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) A comprehensive gas chromatog. anal. for therapeutic concns. of amitriptyline (I) [50-48-6], nortriptyline [72-69-5], imipramine [50-49-7], desipramine [50-47-5], doxepin [1668-19-5], and desmethyldoxepin [1225-56-5] in plasma, is described with use of a N detector. All these drugs are extd. and chromatographed under identical conditions. Each tertiary amine tricyclic is well resolved from its secondary amine metabolite on a mixed-phase column and the concns. of both are detd. simultaneously, without derivatization. The lower limit of sensitivity is 10 .mu.g/L of plasma (2-ml sample). Analytical recoveries of the tertiary and secondary amines are 100 and 80%, resp. Between-run CV's for all of the drugs ranged between 5 and 7%.

Pretreatment with p-chlorophenylalanine [1991-78-2] (400 mg/kg, i.p.) reduced fluid consumption at 24 h, and levels had not returned to normal in 15 days.
