DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) A sensitive, specific and rapid RP-HPLC with UV detection method was developed to det. olamufloxacin (HSR-903) in rat plasma in order to study its preliminary pharmacokinetics in rats. The Mighty RP-18 column (5 mm, 4.0 mm x 150 mm) was used and the mobile phase consisted of ammonium biphosphate and acetonitrile (3:1, pH 2.5) and the detection was at 308 nm. The plasma samples were pretreated using simple liq.-liq. extn. with NR-762 as the internal std. After oral administration of HSR-903 (10 mg kg-1) to rats, their concns. in plasma were measured. The time course of mean plasma concns. was simulated by 3P87 program and fitted to two compartments with first-order absorption kinetic process. The estd. main pharmacokinetic parameters were the following: t1/2(Ka) = 0.33 h, t1/2(VFa) = 0.91 h,t1/2(VFb) = 2.50 h, tmax = 0.90 h, cmax = 1.42 mmolL-1,e,AUC = 5.27 mmol.h L-1. The good linearity was found over the range of 0.1 to 2.0 mmol L-1 and the extn. recovery, accuracy and precision of intra-day and inter-day were all qualified. .
