DOCUMENT TYPE: Patent CA Section: 16 (Fermentation and Bioindustrial Chemistry) An amino acid substrate is reacted with a water-insol. dye-contg. compd.-treated cell homogenate contg. aminoacyl-tRNA synthetase [9028-02-8] to produce peptides or peptide derivs. Thus, Bacillus stearothermophilus UK788 cells (6 kg) were suspended in 2 vols. of 100 mM pH 7.5 Tris-HCl buffer and disrupted. The homogenate was centrifuged to obtain a crude tyrosyl-tRNA synthetase [9023-45-4] ext. The crude enzyme soln. was applied to a column of Martlex Gel Blue-A [76543-98-1] which had been treated with 50 mM Tris buffer contg. 5 mM mercaptoethanol, 2 mM Na EDTA, and 0.1 mM phosphophenylsulfonylfluoride. The column was eluted with KCl in 50 mM Tris buffer. The eluate was concd. and desalted to yield tyrosyl-tRNA synthetase. The enzyme soln. (4.1%) 27, MgCl2 0.4, ATP 0.1 g, L-tyrosine 1 mg, pyrophosphatase 200 units, and dithiothreitol 0.01 mg were dissolved in 200 mL of 20 mM pH 8.0 HEPES buffer. The soln. was stirred at 4° for 15 min and to this was added 4 g L-phenylalanine Me ester. The mixt. was allowed to stand at 30° for 1 day. The mixt. yielded 0.7 mg L-tyrosyl-L-phenylalanine [17355-11-2] Me ester. The control, with enzyme soln. treated on a Sephadex column, yielded 0.1 mg peptide.
