Thus, I, 4-aminonitrefazol [93289-96-4], 4-acetamidonitrefazol [93289-97-5], and p-phenylenediamine [106-50-3] (hydrolysis product of the I metabolite II [140-50-1]) were added to a urine sample which then was acid hydrolyzed and extd. at pH 11. The hydrolyzate ext. was then codetd. with 2-amino-5-chlorobenzophenone [719-59-5], 2-amino-5,2'-dichlorobenzophenone [2958-36-3], 2-methylamino-5-chlorobenzophenone [1022-13-5], 2-amino-5-chloro-2'-fluorobenzophenone [784-38-3], and 2-amino-5-nitrobenzophenone [1775-95-7] by TLC, using toluene as the solvent, and gave no interference. Neither did I and its metabolites interfere in the detn. of 2-amino-5-bromobenzoylpyridine [1563-56-0] by TLC using CHCl3-Me2CO (8:2 vol./vol.) as the solvent system. Thus, the usual methods for benzodiazepine screening may be used for detection of I and its metabolites, using mass spectrometry for identification. Sepn. is also described of I, 4-aminonitrefazol, and 4-acetamidonitrefazol by gas chromatog. on SE 30 or OV 17 on Chromosorb G using N as the carrier gas, and by TLC on DC silica gel plates, using CHCl3-Me2CO, CHCl3-EtOH (8:2 vol./vol.), or toluene-isoPrOH-25% NH3 (:5) solvent systems, with IR and UV spectral data.

The unchanged drug and its metabolites were extd. with benzene-dichloromethane (4:1), subjected to TLC, and detd. by direct UV densitometry. Recovery expts. showed that the method was quant. The limit of detection was 5 ng/mL for 2-amino-5-nitrobenzophenone and 10 ng/mL for other compds. The method was applied to the detn. of nitrazepam and its metabolites excreted in human urine after administration of 10 mg of the drug.
