D(-)-b-hydroxyisobutyric acid (I) [1910-47-0] is produced from isobutyric acid [79-31-2] or methacrylic acid [79-41-4] by fermn. with any of several microorganisms. Thus, Candida rugosa IFO 0750 was cultured in a broth (pH 7.5) contg. glucose 40, yeast ext. 5, peptone 3, meat ext. 1, and isobutyric acid 20 g/L for 24 h with aeration. The pH was raised to 8.5, and fermn. was continued for 48 h. The yield of I was 8.2 g/L.

Thus, C. rugosa was cultivated at 30° with aeration in a pH 7.5 medium contg. cerelose 3.3, yeast ext. 0.3%, and salts. After 23 h, isobutyryl chloride was added and the pH adjusted to 8.35. The broth contained 0.46% I after 43 h of fermn. The broth was clarified, adjusted to pH 2.5, and satd. with (NH4)2SO4. I was extd. from the broth with EtOAc and purified by high vacuum distn. at 106-8°.
