I [R = Me(CH2)n, Me2CH, Me3CCH2, Ph; n = 0-6] were prepd. by acylation of 5'-azido-5'-deoxythymidine, followed by catalytic hydrogenation. All the derivs. showed activity against HSV-1 virus similar to the parent compd., except the arom. and the highly branched aliph. esters (I; R = Me3CCH2, Ph) which were less active. Owing to their markedly improved lipophilicity, these compds. are believed to penetrate the biol. membranes more easily, and thus, would be more effective for the topical treatment of cutaneous herpes virus infections. .
