The most active antisecretory compd. in the pyloric-ligated rat was 2,4,5,6-tetrahydro-1,3,4,6,6-pentamethyl-2-phenylcyclopenta[c]pyrrole-4 -carboxamide (I) [54609-89-1], which gave a pH of 6.2 of the gastric contents when administered orally at 100 mg/kg. I also inhibited histamine-induced gastric acid secretion in the dog, and was only modestly emetic, and did not have anticholinergic properties. Seven other title compds. with small aliph. or alicyclic substituents in the 2 position also were active. Structure-activity relations are discussed. .
