DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) A rapid UV derivatization procedure was developed which allows complete sepn. of PGE1 [745-65-3], PGE2 [363-24-6], PGA1 [14152-28-4], PGA2 [13345-50-1], PGB1 [13345-51-2], and PGB2 [13367-85-6] in <12 min on a C18 reversed-phase column with a CH3CN-10 mM aq. HCOOH gradient as the mobile phase. With pyridinium dichromate [20039-37-6] in CH3CN, the prostaglandins could be completely and selectively oxidized to their corresponding 15-oxo derivs. in 8 min. The oxidized prostaglandins were found to have UV max. at 228 nm (PGE1, PGE2, PGA1, and PGA2) and 298 nm (PGB1 and PGB2). Thus, prostaglandins, which otherwise need to be chromatographed by isocratic elution at 192 nm where the detector noise is considerable, can now be analyzed by gradient elution at 228 nm. Prolonged reaction of PGE1 and PGE2 with pyridinium dichromate gave rise to a 2nd product, probably by oxidn. of the nonallylic 11-hydroxyl group, but within 8 min peak heights were <3% relative to the major product. The smallest detectable amt. was 30-80 pmol. .
