DOCUMENT TYPE: Patent CA Section: 16 (Fermentation and Bioindustrial Chemistry) Section cross-reference(s): 11, 63 A suspension culture of Coleus blumei cells is reproduced in the maintenance medium of Gamborg et al. (1968) contg. 10-30 g sucrose [57-50-1]/L as molasses etc., and used for inoculation of prodn. medium with 1-10 g cells fresh wt./L, equiv. to 30 vol. parts of the maintenance culture/70 vol. parts prodn. medium. The prodn. medium contains 50 g sucrose/L and is fermented from 2 vols. of 75 g sucrose/L plus vol. used and reconditioned maintenance medium. The prodn. culture is aerated and agitated at 50 rpm with a patented stirrer which combines an intensive agitation with low shear for 7 days during which the content of rosmarinic acid [20283-92-5] increases at 0.4 g/L/day to a max. of 3.1 g/L at harvest, followed by lyophilization which yields a light-brown cell powder contg. 16-18 dry-wt.% rosmarinic acid. The lyophilization is used for manuf. of medical prepns. or for increasing the content of rosmarinic acid in herbal prepns.

DOCUMENT TYPE: Journal CA Section: 16 (Fermentation and Bioindustrial Chemistry) Section cross-reference(s): 11 Rosmarinic acid (I) [20283-92-5] formation by intact leaves and suspension cultures of sage and Rosmarinus officinalis varieties was evaluated. In general, cultures produced markedly more I than intact leaves. Marked differences occurred in levels of chorismate mutase [9068-30-8] and phenylalanine ammonia-lyase [9024-28-6] between high- and low-producing cultures of Rosmarinus. The high-producing line exhibited major peaks of both enzymes at ~9-10 days which corresponded to the increase of I during this period, whereas the low-producing line was devoid of these peaks. Optimal levels of I for both lines were attained in the stationary growth phase (12-13 days).
