DOCUMENT TYPE: Conference CA Section: 1 (Pharmacology) Several compds. including Au antiarthritics and related structures were tested as inhibitors of the myeloperoxidase (MPO) [9003-99-0] system of human neutrophils; phosphine Au thiolates were the most effective. Structure-activity studies indicated that the SH group in the mol. was essential for MPO inhibitory activity. Of the compds. tested, the first 4 most effective were SKF-D-100336 (I) [83533-66-8], SKF-D-40079 [83678-72-2], SKF-58889 [51365-22-1], and auranofin [34031-32-8], all contg. Au. Au, however, was apparently not essential although it improved the effectiveness of compds. contg. it such as aurothioglucose [12192-57-3] and aurothiomalate [12244-57-4], compared to identical compds. lacking it such as 5-thio-D-glucose [20408-97-3] and thiomalate [70-49-5]. Inhibition of the MPO system may be one of several mechanisms whereby Au-contg. antiarthritic drugs produce their therapeutic benefit.
