I is cytotoxic to normal mouse fibroblasts (3T3 cells) at 20 mg/mL and to hepatoma cell strains HTC and ZHC at 33 mg/mL. 7b-Hydroxycholesterol (II; R = H, b-isomer) [566-27-8] is cytotoxic to HTC and ZHC cells at 33 mg/mL but not to 3T3 cells at ￡80 mg/mL. 7b-Hydroxy-campesterol (II; R = Me, b-isomer) also shows cytotoxic activity. I and II are probably products of the metab. of the corresponding dietary sterols by B. bassiana.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) D9(11)-Peroxy-5a,8a-cholesterol [78342-39-9], D9(11)-peroxy-5a,8a-ergosterol [86363-50-0], peroxy-5a,8a-cholesterol [14231-33-5], and peroxy-5a,8a-ergosterol [2061-64-5] were synthesized and shown to inhibit 3-hydroxy-3-methylglutaryl-CoA reductase [37250-24-1] activity in cultures of human skin fibroblasts. These sterol peroxides, which have been isolated from tunicates, were similar in their enzyme-inhibiting activities. Inhibition of this early step in cholesterol biosynthesis may be related to the cytotoxicity and potential antineoplastic activity of these oxygenated sterols. .
