DOCUMENT TYPE: Journal CA Section: 62 (Essential Oils and Cosmetics) The oxidative dyes, 4,4'-diaminodiphenylene sulfate [21136-70-9], N'-(2-hydroxyethyl)-4-nitro-o-phenylenediamine [96512-75-3], N,N-dimethyl-p-phenylenediamine [99-98-9], 2,3-dihydroxynaphthalene [92-44-4], and resorcinol [108-46-3], were evaluated for teratogenic potential. Dyes were administered by gavage to pregnant Sprague-Dawley rats on gestation days 6 through 15 at 12.5-500 mg/kg. No overt signs of toxicity were obsd. during the treatment period. A significant redn. in mean maternal wt. gain was noted during the treatment period at the high dose for 4,4'-diaminodiphenylene sulfate, N'-(2-hydroxyethyl)-4-nitro-o-phenylenediamine, and 2,3-dihydroxynaphthalene. The high dose for N,N-dimethyl-p-phenylenediamine and resorcinol, which exceeded a 100-fold exaggeration of human exposure, did not produce a significant redn. in maternal wt. gain. An evaluation of fetal external, visceral, and skeletal anomalies revealed no statistically significant differences between dye-treated and vehicle control groups. Administration of 100,000 units of Vitamin A, the pos. control, on Day 9 of gestation resulted in a significant increase in abnormal fetuses.

DOCUMENT TYPE: Patent CA Section: 25 (Benzene, Its Derivatives, and Condensed Benzenoid Compounds) Benzidine is prepd. by rearrangement of hydrazobenzene, using H2SO4 in a toluene-water system. 21136-70-9 and 92-87-5 which are cas registry numbers are also used here. A mixt. of 1250 g 20% H2SO4 and 400 mL toluene was treated with 180 g hydrazobenzene (prepd. by redn. of nitrobenzene). The benzidine sulfate formed was sepd. by filtration, treated with water and toluene at 90°, followed by addn. of Na2CO3 to pH 7 to give benzidine (99.5% purity) by cooling. The method is environmentally safe. .
