DOCUMENT TYPE: Journal CA Section: 3 (Biochemical Interactions) Each of 6 kynurenines tested (DL-kynurenine sulfate [2126-91-2], quinolinic acid [89-00-9], 3-hydroxyanthranilic acid [548-93-6], xanthurenic acid [59-00-7], picolinic acid [98-98-6], and nicotinic acid [59-67-6]), injected into brain ventricles of mice in doses of 25-50 mg, produced motor excitement and(or) clonic convulsions. Anthranilic acid [118-92-3] did not produce these effects. The strongest metabolite was quinolinic acid, which was active in a dose of 1 mg. It was also the only compd. which produced motor excitement and convulsions after i.p. injection (in doses of 400-600 mg/kg). The hypothermic effect of intraventricularly injected kynurenines was roughly similar to that of i.p. injected kynurenines at 100-1000-fold higher doses. Penetration of kynurenines formed in the liver into the brain appears to be poor. These metabolites of tryptophan (particularly if they are formed inside the brain) may be involved in the mechanism of seizures.
