DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Section cross-reference(s): 28 A series of 2-Me or 2,6-di-Me substituted 1-alkoxybenzyl-4-(pyrimidyl)piperazines I (R = H or Me; R1 and R4 = H or OMe; R2 = R3 = OMe or R2R3 = OCH2O) was prepd. by condensing 2-methyl- [109-07-9] or cis- [21655-48-1] or trans-2,6-dimethylpiperazines [2815-34-1] with 2-chloropyrimidine [1722-12-9] to give the corresponding 1-(2-pyrimidyl)piperazines which were reacted with the appropriate alkyloxybenzyl bromide. 1-(2-Pyrimidyl)-4-(3,4-dimethoxybenzyl)-2-methylpiperazine (II) [97993-28-7] was obtained by condensing 2-methylpiperazine with 3,4-dimethoxybenzyl bromide [21852-32-4] to give 1-(3,4-dimethoxybenzyl)-3-methylpiperazine [97993-29-8] which is reacted with 2-chloropyrimidine, yielding II. None of the compds. had any a-adrenolytic activity. Only 2 of the compds. had dopaminergic activity as detd. by increased mouse motility in reserpinized mice, but the activity of these compds. was not further confirmed by other tests predictive of central dopaminergic activity. II was a more potent vasodilator than piribedil, but such activity did not appear to be correlated with a-adrenolytic or dopaminergic activities. .
