DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Several anthelmintic drugs that are used routinely in oxyuriasis therapy were analyzed for genotoxicity in a diploid mitotic recombination and gene conversion assay (strain D5 of Saccharomyces cerevisiae), and in a haploid yeast reversion assay (strain XV185-14C). Piperazine citrate [144-29-6], piperazine adipate [142-88-1], mebendazole [31431-39-7], and thiabendazole [148-79-8] did not appear to be genotoxic in either yeast strain. Pyrvinium pamoate [3546-41-6] induced the reversion of the missense, nonsense, and frameshift alleles in strain XV185-14C, whereas pyrantel pamoate [22204-24-6] induced only the reversion of the frameshift allele. Pyrvinium pamoate was recombinogenic in strain D5, and there is an indication that pyrantel pamoate, at the lowest dose that was tested, might induce gene conversion or aneuploidy. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Thirteen standardbred horses, all from one farm, were found with a natural infection of small strongyles which were resistant to cambendazole (I) [26097-80-3] when it was first administered to the horses. The small strongyles were found resistant also to 2 other benzimidazole compds. thiabendazole [148-79-8] and mebendazole [31431-39-7]. It is presumed that the strongyles had developed a resistance to the 2 latter compds. which had been used previously and routinely for many years and that the resistance was uncovered when I was 1st used. The small strongyles were susceptible to the following nonbenzimidazole compds: piperazine [110-85-0], pyrantel pamoate [22204-24-6], dichlorvos [62-73-7], piperazine-carbon disulfide complexes with phenothiazine [64568-91-8], and piperazine-thiabendazole mixt. [58428-44-7] and piperazine-trichlorfon-phenothiazine mixt. [64568-90-7]. Some reasons for the development of resistance in nematodes and some considerations for control programs are discussed.
