A study was designed to investigate the hormonal effects of i.v. infusion of isoxsuprine-HCl [579-56-6] (150-200 mg/min) or ritodrine-HCl (I) [23239-51-2] (100-150 mg/min) in a series of 28 women at 28 to 40 wk gestation, with and without concomitant dexamethasone [50-02-2] therapy. I was more potent than isoxsuprine in increasing the circulating levels of cyclic AMP [60-92-4]. The levels of placental steroid hormones decreased slightly, as did the ratio of progesterone [57-83-0] to estradiol [50-28-2]. Human placental lactogen and pituitary hormones showed no consistent changes. Simultaneous administration of dexamethasone did not modify these results.

DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The actions of ritodrine hydrochloride [23239-51-2] which has been effectively prescribed for the prevention of premature labor, on the uterus and heart were compared with those isoxsuprine [395-28-8] and isoproterenol [7683-59-2]. Ritodrine suppressed the spontaneous motility of pregnant rat uterus and showed pos. chronotropic actions in guinea pig atria. In Ca2+-free, K+-rich Tyrode soln., ritodrine suppressed the Ca2+-induced contracture of pregnant rat uterus and potentiated the carbachol [51-83-2]-induced contraction. Ritodrine increased the amt. of cAMP [60-92-4] in the uterus but not in the heart, an action that was suppressed by pretreatment with propranolol. Ritodrine apparently exerts its effects through activation of cAMP prodn., as in the case of isoproterenol, and acts more selectively on b2-adrenoceptors than on b1-adrenoceptors.
