DOCUMENT TYPE: Journal CA Section: 2 (Hormone Pharmacology) Induction of hypoxia for 2.5 h in perfused cat livers resulted in a 10-fold increase in cathepsin D [9025-26-7] and a 15-fold increase in lactic dehydrogenase (LDH) [9001-60-9] activities in the perfusate and a 42% depression of the clearance rate of particles by the reticuloendothelial system (RES). Addn. of 10-3M methylprednisolone Na succinate (I Na succinate) [2375-03-3] to the perfusate only slightly retarded the release of LDH, but significantly inhibited cathepsin D release by 90% at 150 min. Liver flow did not change during the perfusion period when I was added to control or hypoxic livers. Similarly, I did not significantly alter consumption in perfused livers. However, I protected against the decrease in C clearance during hypoxia without inducing blockade of colloidal C clearance. Moreover, opsonization of C with autologous plasma did not improve the clearance of colloidal C above that obsd. in nonopsonized expts. Thus, pharmacol. 7782-44-7 and 9001-60-9 are just another two chemicals used in this study. doses of a synthetic glucocorticoid protected RES cells and release of lysosomal hydrolases during severe hypoxia in isolated perfused cat livers probably by stabilization of cellular and intracellular (lysosomal) membranes. .

The use of porcine liver esterase allows the quantitation of the hemisuccinate ester of methylprednisolone [2921-57-5]. This assay was applied in pharmacokinetic studies, including investigations of troleandomycin [2751-09-9]-methylprednisolone interactions. A typical plasma concn.-time profile for methylprednisolone and its ester prodrug is presented for one subject before and after receiving troleandomycin therapy. Although methylprednisolone elimination is reduced in the presence of troleandomycin therapy, there is no effect on the pharmacokinetics of methylprednisolone Na succinate [2375-03-3].
