DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Toxic side effects of local anesthetics such as procaine [59-46-1], tetracaine [94-24-6], oxybuprocaine [99-43-4], lidocaine [137-58-6], mepivacaine [96-88-8], carticaine [23964-58-1], and bupivacaine [2180-92-9] were assessed by using an in vitro model (suspension culture of Ehrlich ascites tumor cells). All tested local anesthetics at clin. EDs inhibited cell growth; the most pronounced inhibiting effects were obsd. with oxybuprocaine, tetracaine, and bupivacaine. The growth inhibitory effect of procaine pointed to a colchicine-like effect in the metaphase, whereas the inhibitory effect of the remaining compds. appeared to be due to an interference in the interphase of these cells. DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The carbanilate local anesthetics carbisocaine [68931-03-3], heptacaine [55792-21-7], and pentacaine [104485-01-0] liberate histamine [51-45-6] from isolated rat mast cells. Procaine [59-46-1], carticaine [23964-58-1], trimecain [616-68-2], cocaine [50-36-2], and butanilicaine [3785-21-5] were ineffective. Histamine liberation was dose-dependent, followed by calcium displacement from membrane binding sites and occurred without concomitant degranulation. Low temp.- and pH-dependent inhibition of histamine liberation indicated a non-specific, membrane-perturbing effect of highly liposol. carbanilate local anesthetics.
