A common glycosidic structure composed of D-galactose [59-23-4], D-mannose [3458-28-4], and N-acetyl-D-glucosamine [7512-17-6] units was identified which plays a role in insulin receptor interaction and modulates the receptor affinity. The same glycosylation characteristics were found in 2 insulin receptor subunits of mol. wts. 195,000 and 145,000. Pathol. transformation (ascites hepatoma cells) enriched membrane glycoproteins in sialic acid and L-fucose [2438-80-4] residues but did not modify insulin recognition. In cultured hepatoma cells, glycosidic structures were also involved in insulin biol. function, e.g. glycogen [9005-79-2] formation and receptor down-regulation.
