DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Minor tranquilizers (diazepam (I) [439-14-5], nitrazepam [146-22-5], oxazepam [604-75-1], bromazepam [1812-30-2], medazepam [2898-12-6], fludiazepam [60918-52-7], meprobamate [57-53-4]) at low doses increased ambulation score to 145-288% of control rats. Nitrazepam, I and bromazepam increased the ambulation at lower doses to a greater extent than did the other drugs. Fludiazepam and nitrazepam showed a max. increase in ambulation at the same dose. Fludiazepam, nitrazepam and I proved to have potent inhibitory effects on defecation. Trifluperidol [749-13-3], haloperidol [52-86-8] and ID-4708 [50528-46-6] (a new butyrophenone deriv.) and chlorpromazine-HCl [69-09-0] when given at low doses reduced ambulation, while at higher doses defecation was inhibited. These 4 drugs reduced ambulation and restored defecation in methamphetamine treated rats. Clozapine [5786-21-0], thioridazine-HCl [130-61-0] and fluoropipamide-2HCl [2448-68-2] inhibited defecation at nearly the same doses which reduced ambulation in rats not given the methamphetamine treatment. These 3 drugs reduced ambulation, but did not produce a recovery in the defecation rates in methamphetamine-treated rats. Thus, neuroleptics such as clozapine, which rarely induce extrapyramidal side-effects when clin. prescribed, inhibit defecation at nearly the same doses which reduce ambulation. In methamphetamine-treated rats, haloperidol was 31 times more potent than was chlorpromazine in inhibiting activity obsd. with ambulation. This ratio in open-field test was close to the ratio of potency of these drugs as antipsychotic clin. prescribed agents.

DOCUMENT TYPE: Journal CA Section: 64 (Pharmaceutical Analysis) The fluorescence properties of 8 neuroleptic butyrophenones, e.g. benperidol [2062-84-2], were detd. in various solvents and the effects of acidity were examd. Pipamperone [2448-68-2] exhibits a weak signal at all pH but the other ketones show moderate or strong fluorescence. The detection limits of the 4 strongly fluorescing ketones, benperidol, droperidol [548-73-2], azaperone [1649-18-9], and fluanisone [1480-19-9], are ~5 ′ 10-2 mg/mL, after extg. the 4-hydroxybenzoic acid ester preservatives in the liq. prepns. Azaperone fluoresces strongly in acids and has a detection limit of 10-2 mg/mL without removing preservatives.
