Of the systems investigated, DPV at C paste anodes in acetate buffer (pH 5.6) in EtOH-H2O (1:1) is recommended for general anal. work and calibration graphs were linear up to ~1 mM with limits of detection of 4, 20, and 1.5 mM for I, catharanthine, and vindoline, resp. Results obtained by cyclic voltammetry as well as by pulse techniques suggest that electron transfer is preceded by deprotonation and followed by addnl. chem. reactions. Products of the electrode processes form mech. unstable films on the electrode surface.

Thus, 10 kg V. rosea leaves were extd. with 60 L MeOH at ambient temp. for 1 h. After sepn. of the ext., a 2nd extn. with 40 L MeOH followed by extn. with 20L MeOH was used. The MeOH ext. was taken up in EtOAc, the soln. then extd. with 0.5 NH2SO4 and the acidic phase was extd. with hexane. The acidic layer was neutralized to pH 7 with NaOH after addn. of CH2Cl2. The org. phase was dried and concd. at 50° to yield total alkaloid content. The total alkaloids were chromatographed on silica gel to give I and II.
