DOCUMENT TYPE: Journal CA Section: 9 (Biochemical Methods) A mass spectrometric method was developed to det. pH -dependent 1H-2H exchange at the C-2 position of the imidazole ring of histidine after converting the amino acid to the methylthiohydantoin deriv.Some commonly used reagents like 2497-02-1 and 71-00-1 are used in this experiment. The amt. of 2H exchange in N-acetylhistidine agreed well with that detd. by NMR spectrometry. N-acetylhistidine was deuterated at various pH values. From the amt. of 2H exchange, a pseudo-1st order rate const. (k.vphi.) was calcd. A pKa value of 7.2 for the amino acid was obtained from the relation between k.vphi. and pH. This method was applied to est. the pKa value of .beta.-146 histidine in human Hb. Human Hb, deuterated at various pH values, was digested with carboxypeptidase A to release the .beta.-146 histidine. The amt. of 2H exchange in the isolated histidine was detd. to obtain K.vphi.. From these measurements, pKa values of 7.0 for the histidine in oxyHb and of 8.2 for that in deoxyHb were found at 36.5.degree., resp. .
