DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) Drugs were screened for inhibition of 125I-labeled aprotinin [9004-04-0] binding to isolated rat renal brush border membranes. Cationic polymers were effective, and their primary amino groups were crucial. The polycationic aminoglycosides displaced 125I-aprotinin with low concns. (50% inhibition by 50 mg/mL of gentamicin [1403-66-3]). The decreasing sequence in both the no. of amino groups and inhibitory potency was: neomycin sulfate [1405-10-3] > tobramycin sulfate [49842-07-1] > gentamicin > kanamycin sulfate [25389-94-0] > streptomycin sulfate [3810-74-0]. Binding of 3H-gentamicin C1 [25876-10-2] to brush border membrane was saturable. The Scatchard plot indicated an assocn. const. of 43 mM-1, and 18 nmoles per mg of membrane protein for the no. of binding sites. Inhibition of 125I-aprotinin binding by gentamicin was competitive. The inhibition const. (KI) was 20 mg/mL with concns. of 8 and 40 mg/mL of gentamicin. Gentamicin and aprotinin (200 mg/mL) activated b-glucuronidase [9001-45-0] and b-galactosidase [9031-11-2] from renal lysosomes, but not acid phosphatase [9001-77-8]. Gentamicin and aprotinin (300 mg/mL) increased the release of acid phosphatase from intact renal lysosomes. Lysosomal degrdn. of 125I-aprotinin into acid sol. split products was much slower than that of 125I-insulin. Thus, binding to the brush border membrane occurs with chem. quite different, however, basic drugs and the no. of amino groups per mol. is relevant. Nephrotoxicity of aminoglycosides may be related to their endocytic uptake through a direct action on lysosomes.
