DOCUMENT TYPE: Conference CA Section: 1 (Pharmacology) Molsidomine (I) [25717-80-0] i.v. bolus injection at 0.1 mg/kg into conscious, chronically instrumented dogs at rest decreased left-ventricular end-diastolic pressure, contractile vol., and mean arterial pressure, but did not change heart rate, dp/dt, and peripheral resistance. Venous dilation was obsd. 4 min after I administration and reached a max of 22% at 20 min postadministration. The effective compliance of the total vascular system was increased 62% by I. Epicardial coronary artery dilation was obsd. 4 min after I administration and reached a max. of 12% at 20 min postadministration. With i.v. infusion of the I metabolite SIN-1 [33876-97-0], the threshold dose causing epicardial coronary dilation was 0.01-0.03 mg/kg/min, and that causing a decrease in arterial pressure was 3.3 mg/kg/min.

I, however, increased the mean lacate [50-21-5] concn. from 6.9 to 9.2 mg/dL even though conditions of hyperventilation and hypoxemia were not evident. Under conditions of cardiac stress, (90-110 beats/min), I administration decreased the mean lactate consumption when compared to that obsd. in untreated controls; this effect was assocd. with a reduced O consumption by the stressed heart muscle.
