DOCUMENT TYPE: Journal CA Section: 13 (Mammalian Biochemistry) Section cross-reference(s): 6 When the acid-extractable pool of HeLa cells is preloaded by incubation in a high external concn. of an unlabeled amino acid (10-2M) and subsequently exposed to challenge by a physiol. level of a labeled amino acid (10-4M in medium), overshoot occurs, i.e., the acid-extractable pool exhibits a rise in labeling, followed immediately by a fall to equil. At these levels, the height of the overshoot curve is detd. by the preloading amino acid, irresp. of the chirality of the challenge, or whether homo- or heteroexchange is involved. The shape of the overshoot curve is more dependent on the labeled amino acid than on the preloading amino acid. Small differences in the mol. structure of preloading amino acids, such as substitution of F for H, or branching at a different C atom, give rise to as much variation in overshoot as do chirality differences. Preloading with a series of aliph. amino acids shows that the height of the overshoot is correlated with the C-chain length of the preload. A possible mechanism to explain these complicated interactions is discussed. .
