(pH 2.5 adjusted by H3PO4)-methanol-acetonitrile-sodium perchlorate (900 mL:20 mL:80 mL:6 g) as mobile phase, flow rate of 1.0 mL/min, detection wavelength of 220 nm, injection vol. 20 mL and column temp. 40°C. The linear ranges of matrine, oxysophocarpine and oxymatrine were 1.82-12. 26904-64-3 and 519-02-8 are cas registry numbers of chemicals which are used as reagents here.16 mg/mL (r=0.9997), 7.32-48.8 mg/mL (r=0.9996) and 43.45-289.68 mg/mL (r=0.9996), resp. (pH = 2) (7 : 93) at the flow rate of 1.0 mL/min. The HPLC method was performed at column temp. of 30°C, UV detection wave length of 220 nm, and injection vol. of 20 mL. Results showed that the calibration curve was linear (r = 1.0000) in the range of 3.63-116.04 mg/mL (oxymatrine concn.). The mean recovery was 100.1% (RSD = 0.86%; n = 6). In conclusion, this method was effective, and can be used for the detn. of oxymatrine content in oxymatrine injection. .
