Thus, 17.40 lb crude I (byproduct of morpholine manuf.) was heated with 2.1 lb ethylene oxide [75-21-8] at 80°. H2NCH2CH2OCH2CH2NHCH2CH2OH [91598-32-2] was distd. from the mixt. at 180°/0.1 mm in a wiped film evaporator. Curing bisphenol A-epichlorohydrin resin [25068-38-6] with 24 phr this compd. at at 80° for 2 h and 125° for 3 h gave a resin with gel time 23.3 min and Izod impact strength 1.4 ft-lbs/in.

DOCUMENT TYPE: Journal CA Section: 7 (Enzymes) N-Pepstatinyl-N'-dinitrophenyl-1,6-diaminohexane bound nonspecifically to immunoppts. contg. cathepsin D (I). Three new water-sol. localization reagents were synthesized, by using N,N'-bis-(3-aminopropyl)piperazine, 3-oxa-1,5-diaminopentane, or 3,6-dioxa-1,8-diaminooctane as spacer arms between the pepstatin and dinitrophenyl moieties. These dinitrophenyl-pepstatins were bifunctional reagents able to bind the active site of I and anti-dinitrophenyl antibody at the same time. Cultured human synovial cells were fixed and incubated with the dinitrophenyl-pepstatins at pH 5.0 or pH 7.4, then incubated at the appropriate pH value with anti-dinitrophenyl antibody labeled with fluorescein. When examd. by fluorescence microscopy the cells stained at pH 5.0 showed fluorescent perinuclear granules, which were not seen in the cells treated at pH 7.4. The distribution of I detd. by indirect immunofluorescence at pH 7.4, closely resembled that revealed by the dinitrophenyl-pepstatins at pH 5.0. N,N'-(3-Pepstatinylaminopropyl-3'-dinitrophenylaminopropyl)piperazine gave the most intense lysosomal staining and showed no nonspecific binding.Except for chemicals metioned above, 2752-17-2 and 88053-39-8 are also used. This reagent is therefore suitable for the subcellular localization of the active conformation of I. .
