DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) The concns. of progesterone [57-83-0], androstendione [63-05-8], testosterone [58-22-0], and 17b-estradiol [50-28-2] were detd. in the follicular fluid of spontaneously cycling and synchronized [with Oxolven [27806-42-4], pregnant mare serum gonadotropin (PMSG) [9002-70-4], and human chorionic gonadotropin (hCG) [9002-61-3]] Goettingen miniature pigs. Concns. were detd. using RIA at days 0, 1, and 2 in the spontaneously cycling pigs, whereas in the synchronized animals follicular fluid were aspirated at 72 h after PMSG administration as well as 24 h and 36 h after hCG injection. In another series of expts., 5 spontaneous cycling gilts were examd. In addn. to the above hormones, the concns. of 17a-hydroxyprogesterone [68-96-2], dehydroepiandrosterone [53-43-0], and estrone [53-16-7] were detd. in the follicular fluid and follicular wall and in the medium used to culture the follicles for a 24-h incubation period. At day zero of the cycle, estrogen concns. in follicular fluid were higher than other hormones measured. As the time of ovulation approached, estrogen concns. in the follicular fluid decreased and progesterone concn. increased. In contrast to the results in spontaneously cycling animals, follicular hormone concns. of progesterone were always higher than those of other hormones measured. Concns. of 17b-estradiol decline rapidly after injection of hCG. In spontaneously cycling animals, total steroid concns. detd. in the follicular fluid and follicular wall were significantly lower at day 1 of the cycle than at either days 0 or 2. The follicles contained high concns. of androstenedione and testosterone during the immediate preovulatory period. Apparently, during the ovarian cycle, a blockage of estrogen formation occurs and progesterone formation is stimulated immediately afterward. This results in inversion of the intrafollicular estrogen-progesterone ratio. The relatively low dehydroepiandrosterone concns. suggest the D5-steroid biosynthetic pathway is relatively unimportant in ovarian steroid formation in the pig.

DOCUMENT TYPE: Journal CA Section: 2 (Hormone Pharmacology) Daily oral administration of Sa 45.249 (I) [27806-42-4] to 80 cycling heifers, at doses ranging from 6 to 14 mg/day and over varying treatment periods (from 8 to 16 days) resulted in complete estrus inhibition. Irresp. of the dose and treatment period used, heat and ovulations, as judged by the corpora lutea which became palpable on day 15 of this cycle, occurred between day 2 and 9 after cessation of treatment in 73 of 80 heifers. This pattern was confirmed in a pilot field trial. From artificial insemination at the first and second heat after treatment, 16 of 21 treated heifers and 45 of 55 cows were pregnant. There was no difference in cyclic plasma progesterone levels between heifers treated for 14 days with 10 mg I and untreated controls, when treatment commenced either on day 5 or 14 of the cycle, with the exception of the delay in post-ovulatory rise in the latter treatment.
