DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The effect of nootropics on the retrograde amnesia induced by electroshock was studied on a model of the active conditioned escape reaction (pole jumping). In untreated animals the daily application of electroshock for 3 days, immediately after the training, led to a significant retardation of the development of a conditioned escape reaction. The antiamnestic effect of piracetam (I) [7491-74-9] (100 mg/kg, i.p.), which is good also without pretreatment, may still be potentiated, esp. on the 4th day of treatment, by addnl. administration of the drug beginning 4 days before the 1st day of treatment. In contrast to this, meclofenoxate [51-68-3] (100 mg/kg, i.p.) and pyritinol [1098-97-1] (100 mg/kg, i.p.) produce a marked inhibition of the development of the retrograde amnesia only after pretreatment. Without pretreatment, these drugs exert a slight or no effect. The marked antiamnestic effect of methylglucamine orotate [59404-17-0] (225 mg/kg, i.p.) and nicergoline [27848-84-6] (1 mg/kg, i.p.) is not potentiated by an addnl. pretreatment. With dihydroergotoxin [11032-41-0] (1 mg/kg; i.p.), neither treatment regimen was efficacious in the model used. .
