DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The concn. of cinoxacin (I) [28657-80-9] in body fluids and tissues of the urinary tract were studied in 13 patients given 500 mg I twice daily. There was a wide variation in the peak serum concns. (2.8 to 28 mg/mL, mean 16.9 mg/mL); the highest concns. were generally reached within 3 h of administration. A mean peak concn. of 766 mg/mL was found in urine, and the mean cumulative urinary excretion during a 12-h interval was 65%. In bladder and prostatic tissue, the concns. of I were lower than in serum. The concns. in renal tissue showed a considerable variation, but were generally higher than those in serum. No clear relation was found between the concns. in serum and renal tissue. The use of I for the treatment of urinary tract infections is discussed.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) A spectrophotofluorimetric method for the detn. of cinoxacin (I) [28657-80-9] was evaluated. The fluorescence intensity was measured at the excitation wavelength of 345 nm and the emission wavelength of 425 nm. An almost linear relation was found between fluorescence and concn. of I extd. from urine, serum, and renal and prostatic tissue homogenates. The minimal sensitivity of the spectrophotofluorimetric method was 0.10 mg/mL when testing urine and serum. The corresponding figures for prostatic and renal tissue were 0.05 mg/mg and 0.10 mg/mg wet wt. The results of the detns. of I concns. in urine obtained by this method were compared with those found by means of an agar-well diffusion bioassay. A good correlation was found between the results obtined by the two methods. It is concluded that the spectrophotofluorimetric method is suitable for detn. of the I concns. in body fluids and tissues obtained during maintenance treatment in man.
