DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) A simple procedure for the simultaneous detn. of the anticonvulsants oxcarbazepine [28721-07-5], carbamazepine [298-46-4], and 3 of their metabolites (10-hydroxy-10,11-dihydrocarbamazepine [29331-92-8], trans-10,11-dihydroxy-10,11-dihydrocarbamazepine [58955-93-4], and 10,11-epoxycarbamazepine [36507-30-9]) in serum or plasma is described. The alkalinized sample is extd. with EtOAc, the ext. is evapd. to dryness, and the residue taken up with the mobile phase. An aliquot of the resuspended material is injected into the liq. chromatograph and eluted with H2O:MeOH:MeCN (:5 by vol.) on a 5-mm C-18 reversed-phase column. The eluent is monitored at 254 nm. No interference by other anticonvulsants or by endogenous constituents from the sample was obsd. Owing to its good precision, specificity, sensitivity, and selectivity, this method is well adapted to the therapeutic monitoring of oxcarbazepine or carbamazepine treated patients, as well as for pharmacokinetic studies.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) A high-performance liq. chromatog. method for the simultaneous detn. of carbamazepine [50-06-6], CGP 10000 [58955-93-4] (a common metabolite of carbamazepine and oxcarbazepine [28721-07-5]), desmethylmephenytoin [631-07-2], 10,11-epoxycarbamazepine [36507-30-9], ethosuximide [77-67-8], GP 47779 [29331-92-8] (the active metabolite from oxcarbazepine) mephenytoin [50-12-4], phenylethylmalonamide [7206-76-0], primidone [125-33-7], pheneturide [90-49-3], phenobarbital [50-06-6], and phenytoin [57-41-0] is described. The serum is extd. with EtOAc at pH 3.9 and the dried ext. is dissolved in 70% EtOH in water and an aliquot is injected into a Hewlett-Packard 1084 B liq. chromatograph. A reversed-phase (RP-8) column is used with MeCN and water as the mobile phase. The eluted drugs are detected at 207 nm. The recovery of the compds. varies from 76% to 95% with the day-to-day precision between 3.8% and 9.8%. within-day precision between 1.In this experiment, several chemicals are used like 58955-93-4 and 36507-30-9 8% and 5.8%, and run-to-run precision between 1.0% and 2.6%. .
