DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) After oral administration of 14C-labeled valproic acid (DPA) [99-66-1] to rats, for 3-4 days, 80.5% of the total dose was excreted in the urine within 39 days. In addn., 52-66% of the available radioactivity was excreted in the urine within 24 h of the last dose of DPA. The metabolites of DPA in urine were 2-propylglutaric acid [32806-62-5], 5-hydroxy-2-propylvaleric acid [53660-23-4], 3-keto-2-propylvaleric acid [60113-81-7], and dehydrodipropylacetic acid [60218-41-9]. After administration of DPA to humans, 2-propylglutaric acid, 5-hydroxy-2-propylvaleric acid, and dehydrodipropylacetic acid were obsd. in the urine. In addn., in 2 of 8 patients, high amts. of methylmalonic acid [516-05-2] and hydroxymalonic acid [80-69-3] were excreted in the urine and in 4 patients, high amts. of succinic acid [110-15-6], glutaric acid [110-94-1], 2-hydroxyglutaric acid [2889-31-8], and adipic acid [124-04-9] were obsd.
