DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The elimination, metab., and distribution of 14C-labeled total flavanolic oligomers (OFT), vitamin P factors extd. from V. vinifera, was detd. in the rat, as a function of time. After a single oral administration of 50 mg/kg of labeled OFT, about 70 of the administered dose was eliminated in the first 24 hs: 6 of the dose was excreted as 14CO2 in expired air, 19 was eliminated in urine, and 45% in feces. The major urinary metabolites were hippuric acid [495-69-2], ethylcatechol (I) [28930-20-3], and m-hydroxyphenylpropionic acid (II) [621-54-5]. The major fecal metabolite was I. The major biliary metabolites were vanillic acid (III) [121-34-6] and II. The degrdn. of OFT was dependent on the activity of the microflora in the gut. The distribution of radioactivity in rat tissues shows that there are 2 kinds of target tissues for OFT: connective tissues and tissues of the adrenal-thyroid-pituitary axis.
