DOCUMENT TYPE: Journal CA Section: 2 (Hormone Pharmacology) The effects of melengestrol acetate (I) [2919-66-6] (0.5 mg for 14 days starting at day 15 of the estrus cycle) on the cytophysiol. character of the adenohypophysis in 12 Hereford and Angus heifers were studied. A combination of thick-thin sectioning of resin-embedded material was used to aid in specific identification of individual hypophyseal cells. Alterations were not obsd. in the cytophysiol. activity of the presumptive prolactin-, somatotropin-, thyrotropin-, or corticotropin-secreting cells. A marked increase in chromophobes was obsd. in animals given I. Evidence indicated that these chromophobes originated from gonadotropin-secreting cells. I may alter the precise regulatory control exerted by the hypothalamus, resulting in tonic release of gonadotropins from the adenohypophysis.

The 4 S peak seen in all groups (castrate; castrate + estrogen treated; castrate + estrogen and I treated) contained little specific I binding but the 7.5 S peak, seen only in the estrogen treated animal, was specific for I. Competition studies showed the receptor affinities to be I > 5.alpha.-pregnane-3,20-dione [566-65-4] > melengestrol acetate [2919-66-6] > norgestrel [6533-00-2] > deoxycorticosterone [64-85-7]. There was little competition from corticosteroids, 5.beta.-pregnane-3,20-dione [128-23-4], and 17.alpha.-hydroxyprogesterone [68-96-2]. Receptor levels, measured from Scatchard plot anal. of equil. data, were 7.0 (castrate), 45.2 (estrogen-treated), and 10.5 fmol/mg cytosol protein (estrogen + I treated). The assocn. const. was similar in all 3 groups and was .apprx. 5 .times. 10-9M.
