DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Section cross-reference(s): 2 Whereas pretreatment of healthy men with the serotoninergic antagonist Sandomigran (pizotifen) [15574-96-6] (1 mg, orally) inhibited the prolactin [9002-62-4], growth hormone (GH) [9002-72-6], and cortisol [50-23-7] secretory response to metoclopramide [364-62-5] (10 mg, i.v.), pretreatment with the muscarinic antagonist Gastrozepin (pirenzepine) [29868-97-1] (50 mg, orally) decreased only the metoclopramide-induced GH response. Plasma renin [9015-94-5] activity was unchanged by metoclopramide regardless of pretreatment with either antagonist. Aldosterone [52-39-1] secretion was increased by metoclopramide, but this effect was unaltered by the serotoninergic or muscarinic antagonist. Evidently, serotoninergic mechanisms are involved in the response of cortisol and prolactin to metoclopramide, whereas both serotoninergic and muscarinic mechanisms are involved in the GH response.
