Comparison of the calcd. results from the simple diffusion and the exptl. data reveals the presence of the facilitated diffusion of these substances which has been conceived of as a carrier-mediated process. The values of the partition coeff. f for these peptides were in the range 7 ′ 10-4 ￡ f ￡ 200 ′ 10-4. The calcd. f values gave permeabilities, Ps, in lipids between 10-7 ￡ Ps ￡14 ′ 10-7 cm/s. These values were then used to est. the extn. for peptides from simple diffusion alone which vary 0.3-3.5% compared with the exptl. extn. (0.4-12%), indicating the inadequacy of the simple diffusion alone to explain the exptl. data.. As for the carrier-mediated facilitated diffusion process, the activated-complex theory was used. The extn. in this case depends on the maximal rate of transport (Tmax)f and the reciprocal of the affinity const. Kt for the transport of the peptides through the BBB. The (Tmax)f was deduced to be ~0.46 ′ 10-3 pmol/g ′ s and the Kt ~0.35 nM for methionine-enkephalin [58569-55-4], leucine-enkephalin [58822-25-6], glutathione [70-18-8], carnosine [305-84-0], a-MSH [37213-49-3] and MSH release-inhibiting factor [9083-38-9] and (Tmax)t ~10 ′ 10-3 pmol/g ′ s and the Kt ~7 nM for arginine-vasopressin [113-79-1], b-[D-62-alanyl-69-homoarginyl]lipotropin61-69 [92921-61-4], b-[D-2-alanyl-9-homoarginyl]endorphin [72826-17-6], and a-[D-2-alanyl-3-homoarginyl]endorphin [92921-62-5] to explain the obsd. results. The quant. variation of extn. with the concn. of peptide in the brain capillary was also obtained, and the extn. was shown to decrease with increasing concn. of peptides, tending to a small const. value at high concns. Evidently, carrier-mediated facilitated diffusion is important for the transport of peptides across the BBB.
