DOCUMENT TYPE: Journal CA Section: 63 (Pharmaceuticals) Liposome-type artificial red blood cells (ARBC) were prepd. in carboxymethyl chitin (CM chitin) [52519-63-8] soln. by a 2-step emulsification technique by dispersing human hemolyzate as fine droplets in a lecithin soln. in CH2Cl2 to yield a water-in-oil emulsion, and dispersing the resulting emulsion in an aq. CM chitin soln. to give a water-in-oil-water emulsion. Removal of the org. solvent by evapn. from the emulsion gave an aq. suspension of ARBC stabilized with CM chitin soln. The ARBC are spherical, have a narrow size distribution with a mean diam. of 310 nm. The prepn. contains uni- and multilamellar structures and the CM chitin mols. form a mesh-like structure over the entire ARBC surface. ARBC have an O-binding ability similar to that of the human hemolyzate. The degree of O satn. is lower for the ARBC than for the hemolyzate at partial pressures of O >100 mm Hg, while the shape of oxygenation curve and the values of P50 are quite similar for both ARBC and the hemolyzate soln. The disintegration of ARBC increased in the order Na 1-dodecyl sulfate [151-21-3] > 1-dodecylpyridinium chloride [104-74-5] > nonoxyethylene glycol 1-dodecyl ether [3055-99-0]. No disintegration of the ARBC by bovine serum globulins and bovine fibrinogens at their plasma concns. was obsd. Twenty percent of ARBC, however, was disintegrated by bovine serum albumins.
