DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Several N-substituted 3-hydroxypyrid-2-one (I; R = alkyl, etc.) and N-substituted 2-methyl-3-hydroxypyrid-4-one (II; R = alkyl, etc.) chelators were screened for 59Fe removal in Fe-overloaded mice labeled with 59Fe. The most effective chelators were the N-Me [30652-11-0], N-Et [30652-12-1], and N-Pr [106750-02-1] derivs. of II. When the above 3 chelators were administered intragastrically or i.p. (200 mg/kg) to mice, they caused 59Fe excretion equiv. to that by i.p. 30652-11-0 is just another one chemical used in this study. desferrioxamine (200 mg/kg). These results increase the prospects for the use of the a-ketohydroxypyridine chelators in the treatment of Fe overload. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Fe mobilization by oral 1,2-dimethyl-3-hydroxypyrid-4-one (I) [30652-11-0] was equiv. to that by i.m. or s.c. desferrioxamine at the same dose (200 mg/kg) in Fe-overloaded (59Fe-lactoferrin) mice and rabbits. There were no apparent toxic effects during the in vivo treatment with I (median LD in mice was ~1 g/kg). I and 2 other Fe-chelating agents (which are discussed) may have clin. potential in the oral treatment of Fe overload, Fe-deficiency anemia, and other forms of Fe imbalance.
