Inhibitors of DNA, RNA, and protein synthesis in murine bone marrow cells and colony growth of myeloid precursor cells (granulocyte-macrophage colony-forming units) were used as in vitro end points of myelotoxicity. Further, the antiglioma activity of the 4 nitrosoureas on 2 human gliomas in a clonogenic tumor cell assay and the effect of the nonnitrosourea carbamoylators KOCN [661-20-1], chloroethyl isocyanate [1943-83-5], cyclohexyl isocyanate [3173-53-3], ethyl isocyanate [109-90-0], and ethyl isothiocyanate [542-85-8] on granulocyte-macrophage colon-forming units were studied. At equiv. drug exposures, clonogenic glioma cell kill was significant and comparative for 1,3-bis(2-chloroethyl)-1-nitrosourea, 1-(2-chloro)-3-(b-D-glucopyranosyl)-1-nitrosourea, and chlorozotocin; 1,3-bis(trans-4-hydroxycyclohexyl)-1-nitrosourea showed little activity. In contrast, granulocyte-macrophage colony-forming unit toxicity was low with chlorozotocin and 1-(2-chloroethyl)-3-(b-D-glucopyranosyl)-1-nitrosourea and 1,3-bis(trans-4-hydroxycyclohexyl)-1-nitrosourea. Of the isocyanates, bone marrow toxicity was highest with chloroethyl isocyanate and cyclohexyl isocyanate, intermediate with Et isocyanate, and lowest with KOCN and Et isothiocyanate. Evidently, bifunctional alkylation is essential for antiglioma activity of nitrosoureas and myelosuppression is at least partly linked with carbamoylation but structural entities in the carbamoylating isocyanate rather than a quant. degree of carbamoylation det. the degree of potential myelotoxicity.

The relative rate of reaction of compds. with different functional groups was studied. The reaction of CIC with non-protonated groups such as with cyclohexylamine [108-91-8] (a product of CIC reactions with water) or glycine [56-40-6], is much faster than with alc. OH groups or water. Even in highly dild. solns. of ethanol [64-17-5] or water, the amino compds. mentioned are formed in addn. to products of the reactions with cyclohexylamine (dicyclohexylurea (DHH) [2387-23-7]), and glycine (cyclohexylhydantoic acid [33557-91-4]). The following relative rate of reaction with OH groups was obsd: ethanol > water > menthol [1490-04-6] > eugenol [97-53-0]. The rate of reaction is dependent on the polarity of the solvent. New compds. formed are cyclohexylcarbamic acid menthyl ester (I) [91990-71-5] and cyclohexylcarbamic acid eugenyl ester (II) [91990-72-6]. Because of the protonation of the amino group in the acidic range cyclohexylamine was formed at high rates, whereas in the neutral and basic range different amts. of cyclohexylamine and DHH were found. In a water-ethanol-system varying amts. of cyclohexylcarbamic acid Et ester [1541-19-1] were formed in addn. to compds. with water.
