DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Section cross-reference(s): 13 The effect of triflusal (I) [322-79-2], acetylsalicylic acid (ASA)(I) [50-78-2], and of their principal metabolites 2-hydroxy-4-trifluoromethylbenzoic acid (HTB) [328-90-5] and salicylic acid (SA) [69-72-7], alone or combined with dipyridamole (DIP) [58-32-2] and/or PGE1 [745-65-3] on cAMP [60-92-4] levels in washed rat platelets (37° 4 min), has been detd. The effect of triflusal and HTB was significant at therapeutic concns. of triflusal (1 mM) and HTB (0.5 mM). The effect of HTB was always greater than that of triflusal. ASA, at 1 mM and 5 mM, alone or combined with PGE1 was without effect. When 1mM triflusal was combined with 0.1mM DIP1, an increased effect was obtained. ASA, at the highest concn. tested (5 mM), did not modify the DIP-induced increase of cAMP levels.Except for chemicals metioned above, 58-32-2 and 745-65-3 are also used. .

M.; Garcia Rafanell, J. (Fac. Med., Univ. Auton. Barcelona, Barcelona, Spain). Curr. Ther. Res., Clin. Exp., 22(4), 510-21 (English) 1977. CODEN: CTCEA9. DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The effect of triflusal (2-acetoxy-4-trifluoromethylbenzoic acid)(I) [322-79-2] on hemostasis (blood coagulation and platelet aggregation induced by ADP and collagen) was studied in healthy volunteers and patients with latent thromboembolic diseases or with high risk thereof. The healthy volunteers and 1 group of patients were treated only with I (300 mg orally, 30 min before the 3 principal meals). The other group of patients was treated with I (same dose) jointly with Sintrom [152-72-7]. The results show an augmentation of the anticoagulant effect on Sintrom by I and a significant decrease of the platelet adhesiveness and of the potential thrombodynamic index in the group of patients treated with both drugs. A clear inhibitory effect of I on platelet aggregation was obsd. in all groups. The results suggest an interest in I for the prophylaxis and treatment of thromboembolic diseases and in cases of high thrombotic risk.
