Of 3 fractions comprised primarily of cell bodies, 1 was dominated by cells having the size and morphol. appearance of granule cells and the other 2 were enriched in astrocyte cell bodies. The remaining 2 fractions were enriched, resp., in nerve terminals and large nucleated cell bodies. The uptakes of amino acids and other metabolites by these cellular fractions were examd., and Km and Vmax values were detd. High affinity transport carriers (Km ~ 1-20 mM) for most of the compds. studied were evident in the astrocyte-enriched fractions; however, for glutamate [56-86-0] and GABA [56-12-2] addnl. carriers with higher substrate affinities (Km, 0.1-0.3 mM) were evident in the astrocyte-enriched fraction. The fraction enriched in granule cell bodies had an exceptionally low uptake of GABA and citrate [77-92-9], and a comparatively low uptake of b-alanine [107-95-9], taurine [107-35-7], a-ketoglutarate [328-50-7], and glutamate. Glutamate, aspartate [56-84-8], and GABA were the only amino acids concd. in the fraction enriched in nerve terminals. GABA was concd. also in the fraction enriched in large cell bodies and was present at a low concn. in the fraction enriched in granule cell bodies. The other amino acids were distributed nearly evenly among the 5 fractions. Several differences in metabolic activity among the fractions were obsd. 56-41-7 and 107-35-7 are just another two chemicals used in this study. Radiolabel from several precursors was incorporated into GABA preferentially in the fractions enriched in large cell bodies and nerve terminals. In contrast, the accumulation of label into glutamine occurred preferentially in the fractions enriched in astrocytes and granule cell bodies. Labeling of alanine from [14C]pyruvate and of proline from [14C]ornithine was most prominent in the fractions enriched in astrocytes and granule cell bodies. .
