DOCUMENT TYPE: Conference CA Section: 1 (Pharmacodynamics) In pharmacol. studies of alkaloids from Aconitum and Delphinium in lab. animals aconitine (I) [302-27-2], aconifine [41849-35-8], and lappaconitine [32854-75-4] had central nervous, cardiovascular, and respiratory system effects arising from the presence of benzyl ester and hydroxyl groups in their mol. structures. Acetylation of the hydroxyl groups or saponification of the benzyl esters of aconitine or lappaconitine led to sharp decrease in toxicity and cardiac arrhythmic activity. In contrast to aconitine, aconifine, and lappaconitine, aconitine analogs not contg. benzyl esters (acomonine [54191-56-9], karacoline [39089-30-0], talatisamine [20501-56-8], lycoctonine [26000-17-9], delcorine [52358-55-1], and eldeline [6836-11-9]) had a ganglioblocking or curare-like effect, a myorelaxant antidepolarizing effect on nerve muscle transmission, low toxicity, did not produce cardiac arrhythmias, did not stimulate different nerve receptors, and did not greatly affect the central nervous system. Songorine [509-24-0] exhibited psychostimulant and antidepressant properties and in EEG-desynchronizing effect with potentiation of exogenous stimulants. Songorine enhanced the central effects of phenamine and tryptamine and antagonized the hypothermic and sedative effects of reserpine, aminazine, and bulbocapnine.

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) Lappaconitine (I) [32854-75-4] was isolated from the root of Aconitum sinomantanum, which has been used as an anodyne in the northwestern part of China. The analgesic effects of lappaconitine were demonstrated by using the hot plate method in mice and the rat tail-flick test, after i.p. injection of 6 mg/kg. Assayed with the writhing method, the median analgesic dose (ED50) of lappaconitine in mice was found to be 3.5 mg/kg. In the 2-day test, no jumping was obsd. in mice treated with lappaconitine and then challenged with nalorphine. The single-dose suppression test was applied to est. addiction liability of lappaconitine in morphine-treated rats. The change in body wt. was not significantly different from that seen in the control after given lappaconitine at doses of 8 mg/kg. In morphine-dependent monkeys, lappaconitine at doses of 2 mg/kg, which was the maximal tolerated dose, did not suppress the withdrawal symptoms evoked by sudden morphine withdrawal or by s.c. injection of nalorphine (0.5 mg/kg). In other monkeys, lappaconitine were administrated for 53-92 days, and the phys. dependence was detd. During this period no phys. dependence was obsd. No withdrawal symptoms were obsd. when challenged with nalorphine at doses of 4-8 mg/kg. Anesthesia test on rabbit cornea showed that the surface anesthetic potency of lappaconitine was 8 times stronger than that of cocaine. Its local anesthetic activities on sciatic nerve block in mice and intracutaneous wheal test in the guinea pig were found to be 5 times and about equally, resp., as active as cocaine. Lappaconitine was shown to have an antithermic effect in normothermic and pyrexial mice after i.p. injection of 3 mg/kg. In rats, after i.p. injection of lappaconitine 4 mg/kg/day ′ 7 days, a marked redn. in formaldehyde-induced edema of the hind paw was noted. .
