DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Two proteases, Esperase [9073-77-2] and Alcalase [9014-01-1], derived from Bacillus licheniformis and B. subtilis, resp., are used in laundry products. In testing for the prevalence of IgE antibodies to these enzymes in sera among 300 laundry product workers, 2 problems were experienced in the establishment of a reliable radioallergosorbent test (RAST) for these antigens. The 1st problem was the propensity of the allergen, Esperase, to undergo autolysis, suggesting that solid-phase Esperase might also lose reactivity through degrdn. Treatment of Esperase with phenylmethylsulfonyl fluoride [329-98-6] stabilized the enzyme and permitted the synthesis of a stable solid-phase antigen. The 2nd problem was the finding that sera reactive with Esperase in the RAST were also reactive with Savinase [79986-26-8], an enzyme from B. licheniformis to which the workers were not exposed. Immunochem. analyses of the 3 enzymes with specific rabbit antisera by gel diffusion and by 2-site immunoradiometric assay demonstrated that they were not cross-contaminated to any appreciable extent. RAST inhibition demonstrated that solid-phase Esperase possessed unique allergenic determinants in that the reactivity of IgE antibodies was inhibited by low concns. of Esperase and only by very high concns. of Alcalase and Savinase. In contrast, the reactivity of solid-phase Alcalase was occasionally inhibited equally well by Esperase and Alcalase. Most strikingly, the reaction of IgE antibodies with solid-phase Savinase was always inhibited by comparable quantities of Esperase, Alcalase, and Savinase. Thus, the establishment of the RAST for these proteases appears to require the use of phenylmethylsulfonyl fluoride to retard autolysis, and the results must be interpreted with caution because IgE antibodies in certain sera demonstrate cross-reactivity with Alcalase and Savinase.

DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Phenylmethylsulfonyl fluoride (PMSF) [329-98-6] administration to rats was effective in partially preventing liver necrosis induced by thioacetamide [62-55-5], dimethylnitrosamine [62-75-9], or galactosamine (I) [7535-00-4], when given 6 h after the hepatotoxins. In the case of I but not of the other necrogenic chems., protection was also obsd. when PMSF was given 10 h after this compd. Participation of protein degrdn. at late stages of liver injury by these chems. was suggested.Except for chemicals metioned above, 62-55-5 and 62-75-9 are also used. .
