DOCUMENT TYPE: Journal CA Section: 3 (Biochemical Interactions) Section cross-reference(s): 2 When spermidine trihydrochloride [334-50-9], putrescine-2HCl [333-93-7], or 1,3-diaminopropane [109-76-2] was injected (12.5 mmol/100 g) into rats 1 h before thyrotropin [9002-71-5], ornithine decarboxylase [9024-60-6] activity was increased by 75-150% over control levels. However, when 375 mmol polyamine/100 g was injected, thyrotropin-activated activity was inhibited by 70-95%. Multiple polyamine injections inhibited goitrogen-induced activity and gland wt. increase by ~35%. The polyamines also inhibited thyrotropin-activated rat thyroid ornithine decarboxylase in vitro in a dose-related fashion, with 50% inhibition occurring at 2-5 ′ 10-4M. No stimulation was seen with low concns. of polyamine. The polyamines had no effect on in vitro thyroid protein/RNA synthesis or glucose oxidn. but had a biphasic effect on plasma membrane adenylate cyclase [9012-42-4] activity. A protein inhibitor to thyroid ornithine decarboxylase was generated in vivo by multiple injections of the polyamines into rats, and in vitro by incubating bovine thyroid slices with 2-10 mM polyamine. The inhibitor was nondialyzable, was destroyed by boiling, and its formation was blocked in a dose-related fashion by cycloheximide. Thyroid ornithine decarboxylase is subject not only to pos. control, but is apparently also neg. regulated by its end-products, the polyamines, which induce a protein inhibitor to ornithine decarboxylase.

The waking state was increased, and the non-REM sleep and REM sleep was markedly decreased immediately after the injection of 40 mg spermine tetrahydrochloride (SPM) [306-67-2] to the 8th h, and from the 24th to the 32nd h. The amt. of total sleep was shortened from the 72nd to the 80th h following injection of 40 mg SPD. The administration of 40 mg SPM reduced the amt. of total sleep. A marked change was obsd. in the sleep-wakefulness cycle in the 24th to the 32nd h following injection of 100 mg putrescine-2HCl (PUT) [333-93-7]; non-REM sleep and total sleep were decreased. Thus, the intraventricular administration of polyamines including PUT inhibits the amt. of normal sleep.
