DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) Synthetic gonadoliberin (GnRH) [33515-09-2] (10-10-10-9M) stimulated gonadotroph differentiation in adenohypophyseal primordia explanted from 11-13-day-old rat embryos but only in the presence of insulin [9004-10-8]. 9004-10-8 and 10-10-1 which are cas registry numbers of chemicals are mentioned. A 2nd application of GnRH before the end of the culture period stimulated the release of gonadotroph secretory products. Addnl., endogenous GnRH was detected in fetal pituitary glands removed at 14-21 days of gestation. The material is unlikely to be of maternal origin since very small amts. of radioactivity were detected in 11-day-old fetuses after 125I-GnRH injection to pregnant rats. Apparently, GnRH, in synergy with insulin, is implicated in gonadotroph differentiation in early fetal life and the GnRH is probably of fetal or placental origin. .

Relative potencies of chicken LH-RH were 2.5% that of mammalian LH-RH for LH and 3.2% that for FSH. On the other hand, the ED50 for chicken LH-RH for LH release from chicken anterior pituitary cells was 1.03 nM, indicating that the biol. potency of chicken LH-RH acting on chicken pituitary cells was higher than that of chicken LH-RH acting on mammalian pituitary cells. The ID50 of chicken LH-RH to inhibit the binding of 125I-labeled [D-Leu6]LH-RH analog to rat anterior pituitary membrane was 200-fold that of mammalian LH-RH. Substitution of arginine residue of mammalian LH-RH for glutamine residue of chicken LH-RH apparently causes a decrease in the binding affinity of the hormone for the mammalian LH-RH receptor, suggesting that the mammalian LH-RH receptor has a binding site for the cationic center of the arginine residue of LH-RH. On the other hand, chicken pituitary LH-RH receptor seems to have a broader specificity than mammalian LH-RH receptor. .
